The Du Roc House stood 175 feet due south on the road from Sacramento to Placerville. Beginning on April 3, 1860, the Pony Express used this route between San Francisco and St. Louis, Missouri. The westbound Pony Express stations were Sportsman Hall, El Dorado by way of Placerville and Diamond Springs, and Mormon Tavern. Because of the Civil War, the U.S. Post Office took over the prior private Pony Express operation and on July 1, 1861, made Placerville the western terminus of its Pony Express. To continue to San Francisco, Wells Fargo and Co. operated its own Pony Express by way of Folsom from Placerville and then used the railroad to Sacramento. The Du Roc House became the new Pony Express station from Placerville to Folsom.
