Tradition states that the church was founded in 1723, seventeen years before the Moravians arrived in Nazareth, and although that date has not been proven, Emmanuelsville was clearly among the region's earliest church congregations. The oldest legible grave marker is that of Nicholas Heil, dated February 14, 1760, and the oldest existing church record book dates to 1765. A deed from 1765 mentions both a church and a schoolhouse on the property. By 1776, the original church building had become so dilapidated that services could no longer be held there, and from 1776 to 1850 services were held in the schoolhouse. The oldest part of the present church structure was built in 1850. Half of the financing for the pipe organ installed in 1910 was provided by Andrew Carnegie. Until 1969, the church served both Lutheran and Reformed congregations, and it is now home to Emmanuels Lutheran Church.
